We investigated the changes in gene expression upon reprogramming of mouse embryonic fibroblasts. For this we analyzed large number of data sets from three mammalian species, mouse, chimpanzee and human. A set of nine splicing factors has consistent changes that were observed across the three species. We found that ESRP1 and ESRP2 accelerated and enhanced reprogramming efficiency by inclusion of CD44 epithelial specific isoforms which also enhanced mesenchymal to epithelial transition.overall design: Mouse embryonic fibroblasts from TNG_MKOS mice (Chantzoura et al., 2015. Stem Cell Reports. 2015; 5: 350–364) were overexpressed with ESRP1 and ESRP2. After 4 hours of infections with retroviral particles expressing ESRP1 and ESRP2, reprogramming was induced by adding doxycycline which induces the expression MKOS factors. For the first 5 days of reprogramming, mOrange positive cells and for day 15 Nanog-GFP positive cells were sorted and mRNA-seq samples were prepared as mentioned in Mittal et al., Nat Commun. 2017; 8:457.
