We report the high-throughput profiling of p300 and autoacetylated p300 (acp300) in mammalian cells. By obtaining over two billion sequence reads from chromatin immunoprecipitated DNA, we generated genome-wide occupancy maps for p300 and acp300 in a doxycycline-inducible p53 expression system in H1299 non-small cell lung carcinoma cells. We find that p300 and acp300 have a distinct binding pattern upon p53 expression. The acp300 has a distinct TSS binding pattern while the p300 protein exhibits a stronger signature at the distal/ putative enhancer regions. The ChIP-seq data suggests that the phenomenon of factor-induced p300 autoacetylation may play a pivotal role in the integration of stimulus-driven transcriptional pathways, where the recruitment of the catalytically active form of p300 may trigger a quick transcriptional response to the internal and external signaling cues.
