For single cell RNA sequencing dataset, the 10× Genomics platform was used to perform massive single cell mRNA profiling from the lateral walls of the lateral ventricle (Enface) of adult mouse. Only male C57BL/6 mice were used in the analyses, whole mount enface tissue were dissociated into single cells through our proprietary method developed based on published studies1. Ependymal cells with high CD133 protein expression were enriched by soft Fluorescence-activated cell sorting (FACS) using the WOLF cell sorter from Nano Collector Inc.. CD133 positive cells were labeled as “PE” cells and CD133 negative cells were labeled as “NC” cells. Both “PE” and “NC” cells were subjected to single-cell transcriptomic sequencing using the 10× Genomics platform. [1] Liu J, Wang J, Xu J, et al.
