Our findings demonstrated that BCG treatment significantly reduced the intracellular viral titers of Vesicular Stomatitis Virus (VSV). To explore the molecular mechanisms responsible for the enhanced antiviral immunity of BCG-trained macrophages, we conducted RNA sequencing analysis on RAW264.7 cells infected with VSV, comparing macrophages pre-exposed to BCG with those without prior exposure. To explore the impact of BCG-induced trained immunity on antiviral responses, we initially established an in vitro cellular model by treating RAW264.7 cells with BCG lysate for 24 hours, followed by a 5-day rest period. Subsequently, both PBS-treated and BCG-trained RAW264.7 cells were infected with Vesicular Stomatitis Virus (VSV) in vitro for 48 hours.
