To investigate whether the over-expression of c-Myc gene can sensitize cancer cells of multiple tissue origins to Tipifarnib. We performed differential gene expression analysis using RNA-seq profile of three c-Myc over-expressed and control cell lines with/without Tipifarnib treatment. Overall design: We over-expressed c-Myc in three cancer cell lines without c-Myc amplification, SKOV3, A549, and MDA-MB-231, representing ovarian, lung, and breast cancer, respectively, using lentivirus. The over-expression cell lines and three control cell lines were treated with Tipifarnib or DMSO as control for 48 hours before RNA extraction. Differential gene expression analysis was performed using the RNA-seq data to identify genes associated with the change of Tipifarnib sensitivity between the c-Myc over-expressed cell lines and the control cell lines.
