DCC1 knockout in yeast and human cells results in cohesion and DNA replication defects. These findings suggest that loss of Dscc1 in mammalian cells should be incompatible with embryonic development, yet we have generated a Dscc1-null mouse. Primary cells derived from Dscc1-null mice exhibit the replication defect but show normal cohesion. To ascertain whether cohesion is rescued by mutation of a âbypassâ gene, we wish to exome sequence a number of Dscc1-null mice.
