Discovery of novel auxin-responsive genes of Arabidopsis using auxin inhibitors. Arabidopsis thaliana

To obtain more information on auxin-regulated gene expression, we treated Arabidopsis seedlings with auxin biosynthesis or signaling inhibitors, then performed DNA microarray analyses. Overall design: Arabidopsis Col-0 seedlings were grown on 1/2MS agar medium containing 1% sucrose for 6 or 7 days. The seedlings were transferred into 1/2MS liquid medium containing 1% sucrose and grown with constatnt shaking for 1 day, and then treated with IAA and auxin inhibitors for 3 hours. Total RNA was extracted using RNeasy Plant mini Kit (QIAGEN).
