The goal of this study was to perform transcriptomics of prostate cancer cells with genetic knockout and rescue of OGDHL treated with the Androgen Receptor Pathway Inhibitor Enzalutamide (Enza) or Vehicle (DMSO) Overall design: Prostate cancer cells were infected with lentivirus containing CAS9 and a gRNA targeting OGDHL (sgOGDHL). Cells were then maintained in Androgen Receptor Pathway Inhibitor (Enza) for 6 weeks to adapt. Knockout cells were then infected with lentivirus carrying control enhanced green fluorescent protein (eGFP), knockout-resistant full-length (FL) OGDHL, or knockout-resistant deleted mitochondrial targeting sequence (dMTS) mutant of OGDHL.
