In this project, we present a UMI-based small sub-unit rRNA (e.g. 16S rRNA) amplicon sequencing method, termed ssUMI, that is both high-throughput and highly-accurate on the Nanopore platform using Q20+ chemistry, and showcased the high-throughput scalability of this method by sequencing 90 wastewater samples on a single PromethION flowcell for accurate microbial profiling.
