The study aims to investigate functions of PfNOT1.1 in regulating mRNA decay in the malaria parasite, P. falciparum. To achieve this goal, transcription was blocked by actinomycin D (actD) and the transcriptional profilling of parasites was analyzed by microarray technology. Overall design: The endogenous PfNot1.1 was disrupted. The transgenic line and the wildtype line were treated with actD to block transcription or DMSO as a control for four hours. Transcriptional profiling of initial parasites at 24 hpi, parasites treated with actD and parasites treated with DMSO was assessed by microarray.
