PoxTMA7 containing a highly conserved domain TMA7 (Translation machainary associated protein 7). The deletion of PoxTMA7 significanlty reduced the cellulase production by 70% in P. oxalicum, but the underlying translation regulatory mechanism remain unclear. In order to inveatigating the mechanism of PoxTMA7 in translation regulation. We performed ribosome profiling sequencing (Ribo-seq) of WT and detaPoxTMA7 to explaining the landscape of PoxTMA7 in translation regulation. Overall design: Mycelia of P. oxalicum strains WT and detaPoxTMA7 were collected after cultured in medium containing 1% Avicel, and then powdered in liquid nitrogen. Powdered mycelia were mixed with cold polysome extraction buffer, and lysed on ice for 25 min. The ribosome protected frames were extracted by Trizol method after RNase I digested. he Ribo-seq library construction of P. oxalicum was measured by QEZ-seq 1.0 kit.
