Transcriptomic analysis of REVOLUTA and mir165a cell-types of Arabidopsis shoot apical meristem

Dorso-ventral (DV) patterning at the Shoot Apical meristem (SAM) is essential for organogenesis in Arabidopsis. Dorsal and ventral gene expressed domains are separated by boundaries from where the new organ initiation starts. Here by using cell-types specific transcriptomics approach, we have identified dorsal, ventral and boundary specific genes in SAM. 6 different cell-types, and 4 biological replicates for each cell-type were collected through FACS, and used for RNA-Seq analysis. The corresponding position for each cell-type is as following: BFPYFP- Epidermal Rev expressing cells, BFPGFP- epidermal miRNA165a expressing cells, BFP- Epidermal cells not expressing REV and mir165a, YFP-subepidermal cells expressing REV, GFP-subepidermal cells expressing miRNA165a, Allneg-Cells not expressing any marker.
