Most RNA-seq studies on ejaculates of mammalian species do a previous purification step to isolate spermatozoa and remove somatic cells. Often this process is done by gradient centrifugation. However, this process requires fresh samples and limits the origin and the throughput of sample processing. We aimed at comparing the RNA-seq transcriptome profiles of purified and the paired unpurified samples from 4 porcine ejaculates. Our results show high correlation between purified and paired unpurified samples with few genes showing differential RNA abundance thereby indicating the RNA purification may not be needed.
