We applied laser capture microdissection (LCM) to precisely dissect single tumor nests from acral melanoma with high cellular purity, each yielding 20-50 cells; subsequently we performed whole genome amplification using multiple annealing and looping based amplification cycles (MALBAC), in order to generate sequencing libraries from the minute amount of DNA extracted from each dissected region.
