Gene expression profile of tumour-infiltrating immune cells in murine PDAC

We established an organoid-based murine pancreatic cancer model that closely resembles human PDAC. To characterize the tumor-infiltrating immune cell states following combination therapy with immune checkpoint inhibitors (ICIs) targeting PD-1 and CTLA-4, along with agonistic CD40 antibodies (aCD40), we performed RNA sequencing (RNA-seq) on tumor-infiltrating CD45+ cells from our model. Overall design: Tumor-infiltrating CD45+ cells were isolated by FACS from tumor-bearing mice treated with either control antibodies or a combination of ICIs and aCD40. These cells were then subjected to RNA-seq analysis, with biological tetraplicates for each group.
