Here, we sought to evaluate the impact of KrasG12D and KrasG12C inhibitors on pancreatic cancer cells. A murine pancreatic cancer cell line with KrasG12D mutation was treated with MRTX1133 (KrasG12D inhibitor) and MRTX849 (KrasG12C inhibitor) and scRNA-seq performed to evaluate transcriptional changes. We further evaluated transcriptional changes in cancer cells, as well as in the tumor microenvironment of immunodeficient (NSG) and immunocompetent (C57BL6/J) mice orthotopically implanted with KPC689 cells and spontaneous KPPC tumors in response to MRTX1133 and MRTX849. KPC689 cells: untreated n=1, vehicle (DMSO) n=1, MRTX1133 (50 nM) n=1, MRTX849 (50 nM) n=1. C57BL6/J and NSG KPC689 tumors: VehicleMRTX1133, MRTX1133, VehicleMRTX849, MRTX849 n=1 sample per group, 3 pooled mice per sample for VehicleMRTX1133, VehicleMRTX849, MRTX849 and 6 mice pooled per sample for MRTX1133 and healthy pancreas. KPPC vehicle: n=1 mouse, KPPC MRTX1133: n=1 mouse
