Plant metabarcoding from sedaDNA of Siberian and Alaskan lake sediment cores

The dataset contains raw files from NGS sequencing using Illumina devices. In total, eight data sets containing forward (*.R1.fastq.gz) and reverse (*.R2.fastq.gz) files are provided from amplicon sequencing of sediment samples from lake sediment cores (sedaDNA). Therefore, 3 to 5 g of sediment was used in the PowerMax® Soil DNA Isolation Kit (Qiagen, Germany) resulting in 504 sedaDNA extracts. The sedaDNA was concentrated and purified with the GeneJET PCR Purification Kit (ThermoFisher), then PCR replicates using the trnL g and h primers were performed and PCR products were purified using the MinElute PCR Purification Kit (Qiagen). Finally, the PCR replicates were pooled equimolarly and sequenced in paired-end mode (2x150bp) on a HiSeq or NextSeq Illumina platform run by the Fasteris Genesupport SA sequencing service (Switzerland).
