Single-nucleus transcriptomics of Parp7-/- lung

To explore the cell type-specific effects of PARP7 deficiency in the lung, we performed single-nucleus RNA sequencing (snRNA-seq) of lung from WT and Parp7-/- lung. Overall design: 10-month-old wild-type and Parp7-/- mice (2 mice/genotype) were euthanized and lung were collected and snap frozen in liquid nitrogen. Nuclei were isolated from frozen lung and droplet-based snRNA-sequencing was performed using 10x Genomics.
