The main objective of the project is to clarify the role of ruderal vegetation, its rhizosphere, endorhizosphere, indigenous microorganisms and bacteriophages in the processes of natural, spontaneous bioremediation of long-polluted soils. Both soils and plants will be taken from underneath oil extracts at the site of the historic Oil Mine. These areas have been heavily polluted and degraded (over 100 years of pollution). The area is richly covered with relict vegetation. DNA will be isolated directly from the soil, rhizosphere and endorhizosphere. Isolation and characterization of bacterial and fungal strains isolated from the rhizosphere and endorhizosphere of ruderal plants will be performed. The strains will be evaluated by morphological, biochemical and genetic tests. The following assays will be performed: next-generation sequencing (NGS) of variable regions (16S rRNA for bacteria and ITS for fungi) and viral NGS (Shotgun sequencing).
