A human kinome CRISPR knockout screen in H358 parental and sotorasib-R H358 cells using Addgene pooled libraries (#75314). Overall design: Parental cells were cultured with DMSO, while resistant cells were cultured with sotorasib for 2 weeks. Integrated sgRNAs were amplified from genomic DNA and sequenced. We analyzed depleted sgRNAs in sotorasib-R H358 cells compared to reference or parental cells.
