Manganese is an essential trace element for plant growth and development, participating in crucial processes such as photosynthesis, protein synthesis, lipidsynthesis, enzyme activity, and defense against oxidative stress. As a vital componentof the photosystem II (PSII) oxygen-evolving complex in plants, manganese isindispensable for photosynthesis. However, other functions of manganese withinchloroplasts remain unclear. To uncover unknown manganese-associated activitieswithin chloroplasts, our laboratory performed EMS chemical mutagenesis on theArabidopsis manganese uptake-defective mutant nramp1 to establish a mutant library. We screened and identified a series of manganese-deficient sensitive mutants namedmdg (Mn-dependent defective growth). This study conducted phenotypic andphysiological analysis on two new yellow leaves mutantmdg5nramp1, as well as cloning and functional analysis of the target genes.
