Effect of Foxo1 deficiency in naive CD4 T cell activation in vitro

Foxo1 deficiency promotes T-cell exhaustion. Naive CD4 T cells from peripheral lymph-nodes of mice deficient (C57BL/6 Foxo1TKO) or proficient (C57BL/6 Foxo1Ctrl) for Foxo1 expression in T cells were purified and 50 000 cells were cultured 4 days in the presence of an anti-CD3 antibody (145-2C11, 0.1 µg per ml) and 250 000 antigen presenting cells (splenocytes from C57BL/6 CD3eKO mice).
