The outdoor-grown dwarf petunia plants (Petunia hybrida) used for sequencing originate from the Flower Practice Teaching Base at Huazhong Agricultural University. The DNA utilized for genome sequencing is extracted from dwarf petunia plants with heavy petals. Preliminary work includes the correction and assembly of non-redundant full-length transcripts. Simultaneously, RNA is extracted from various parts of the dwarf petunia, such as roots, stems, leaves, and flowers, as well as different single-petal varieties, to construct distinct sequencing libraries, resulting in high-quality second-generation transcriptome data.The genome annotation of the cultivated dwarf petunia species primarily references the MAKER gene annotation process adopted by multiple published genomes. This process encompasses repetitive sequence analysis, gene prediction (including evidence from ESTs, RNA-seq, homologous proteins, and ab initio gene predictions), and functional annotation of the sequenced genome post gene prediction. The final annotated results are obtained through the utilization of software, including Hisat2, Cufflinks, RepeatMasker, Augustus, SNAP, MAKER, Blast, among others.
