We developed RELATe (restriction/ligation coupled with Agrobacterium mediated transformation) method to generate sgRNA library from Cryptococcus neoformans genomic DNA, targeting over 98% of the protein-coding genes. High-throughput screen identified 142 potential targets contributing to blood-brain barrier penetration. We validated two targets, SFP1 and WDR1, for our proof of concept demonstration that RELATe is suitable for genome-wide high-throughput screen in fungi and other eukaryotic species for elucidating functional genomics.
