An array of G-quadruplexes (G4s) were inserted in an isolated locus, with low chromosomal interactions and devoid of inherent G4 forming motifs, in the human genome in HEK293T cells. Another similar sequence with specific substitutions which would prevent G4 formation was also inserted at the same locus as a negative control. Hi-C was done in these pair of cell lines to understand G4 dependent looping interactions and the role of G4s as enhancer elements.
