Abnormally activated neutrophils, also known as polymorphonuclear myeloid-derived suppressor cells (PMN-MDSCs), play essential roles in shaping an immune-suppressive tumour microenvironment that creates a major obstacle to effective immunotherapy. Deciphering the key molecules that govern PMN-MDSCs is required to selectively target these cells for improving the efficacy of immune checkpoint blockade (ICB). Here, we utilized CUT&Tag-seq, RNA-seq to identify BRPF1 as a novel epigenetic regulator that induce PMN-MDSCs infiltration and confer anti-PD1 resistance in hepatocellular carcinoma (HCC).
