Exosome is the main RNA surveillance and degradation pathway during oocyte growth. To figure out specific roles of RNA surveillance in oocyte growth, the pathway was destroied by deleting MTR4, the only known RNA helicase in exosome pathway. Accumulation of waste RNAs in nucleus seriously effect chromatin status and establishment of histone modifications like H3K4me3. Thus CUT&Tag experiments targeting at H3K4me3 were performed. The H3K4me3 CUT&Tag was performed with 120 oocytes per sample. WT and MTR4-null oocytes were extracted and duplicates were conducted. Each sample was sequenced 8G.
