Effect of encapsulating peritoneal sclerosis modeling on mice peritoneums

Encapsulating peritoneal sclerosis (EPS) is a life-threatening fibrotic condition characterized by severe abdominal adhesions, chronic inflammation, and significant morbidity. The lack of effective treatments for EPS stems from a limited understanding of its underlying mechanisms. Previous EPS models relied on intraperitoneal injection of surgical hygiene solution (SHS), requiring 8 weeks to establish. Here, we modified the model by integrating both PD and lipopolysaccharide (LPS), leading to the formation of stable adhesions in just 3 weeks.This model better simulates the pathophysiological progression of EPS by reflecting more clinically relevant risk factors, such as PD and peritonitis, and captures key features like inflammatory infiltration and extracellular matrix (ECM) deposition. Overall design: C57BL/6J male mice, aged 8 weeks,were received daily intraperitoneal injections of 100 mL/kg of 4.25% peritoneal dialysis solution (PDS) and 0.3 mL SHS, along with a single weekly intraperitoneal injection of 4 mg/kg lipopolysaccharide (LPS). And mice received the same volume of saline were used as controls. Mice were sacrificed in the 3rd week, when significant intra-abdominal adhesion formation was observed in the EPS modeling mice.
