Data availability file: CellTiter-Blue® assay of hiPSC-CMs monoculture (100% hiPSC-CMs) and co-culture with HCAECs (90% hiPSC-CMs + 10% HCAECs and 80% hiPSC-CMs + 20% HCAECs) in 2D versus 3D microenvironment, at the end of 14 days of culture. 2D system: viability values from co-culture settings were normalized on the hiPSC-CMs monoculture one. 3D system: viability values from co-culture settings were normalized on the hiPSC-CMs monoculture one. Biological replicates =2 and Technical replicates=3. Statistical analysis: 2-way ANOVA (post-hoc Tukey correction) (** p< 0.01; *** p< 0.001). Relative mRNA expression levels of: cardiac maturation structure genes TNNT2, ACTN2, MYL2, MYH7; Gap junction gene CX43/GJA1; gene of metabolism PPAR-a in hiPSC-CMs monoculture (100% hiPSC- CMs) and co-culture with HCEACs (90% hiPSC-CMs + 10% HCAECs and 80% hiPSC-CMs + 20% HCAECs) in 2D (blue bars) and 3D (red bars) microenvironment. The mRNA expression data were normalized by the geometric mean of the most stably expressed genes (RPL13a, RPS4X, and PPIA), and the relative quantification was performed by the ∆∆Ct method. Biological replicates = 2 and techincal replicated = 3. Statistical analysis: the Fisher's test was used after ANOVA and the results were expressed as mean ± SEM (p-value < 0.05 was considered significant). Statistical analysis: parametric t-test (*p<0,05) high sensitive Troponin I (hsTnI) detection assay of hiPSC-CMs monoculture (100% hiPSC-CMs) in 2D microenvironment versus co-culture with HCAECs (80% hiPSC-CMs + 20% HCAECs) in 3D microenvironment, at the end of 14 days of culture. Biological replicates = at least 3 and Technical replicates=3. Statistical analysis: non- parametric t-test (Mann-Whitney test) (**p<0,01). Live-dead quantification analysis of hiPSC-CMs co-cultured with HCAECs (80%-20%) in the GelMA 3D microenvironment. Comparison of normalized value of live cells (green fluorescence) versus dead cells (red fluorescence). Statistical analysis: unpaired t-test (**p<0,01).
