Data and code for High-Dimensional Mediation Analysis (HDMA) in diversity outbred mice

We used a novel method of high-dimensional mediation to infer a composite transcript that mediated the effect of genetics on metabolic traits in a genetically diverse population of mice maintained on a high-fat high-sugar diet to model diet-induced metabolic disease and obesity. We show that transcripts with high distal heritability contribute more to trait variability than transcripts with high local heritability. We also demonstrate the interpretability of the composite transcript and multiple applications for translation and hypothesis generation.
