The most typical swamping wetland, WuLiangSuHai, was referred as a case study combined with landscape spatial heterogeneity focusing on the key microbiological processes of methane oxidation and ammonia oxidation using modern molecular techniques. The methane-oxidizing bacteria and ammonia-oxidizing bacteria community structure, distribution and dynamics will be deeply analyzed, especially the diversity and richness of key functional genes related to these microorganisms; the relationship between methane-oxidizing bacteria and ammonia-oxidizing bacteria (including aerobic and anaerobic, bacteria and achaea) will be clarified, and mechanisms these functional microbe's responses to the environment during the process of swamping will be illuminated; the rates of methane and ammonia oxidation and the formation of methane and nitrous oxide will be tested to quantitatively assessment of its roles in controlling greenhouse-gas emissions. It is hoped that we can reveal reasons caused higher spatial heterogeneity of methane-oxidizing bacteria and ammonia-oxidizing bacteria, and the mechanism combined these microorganisms with its environment functions from diversity of substrates, microbial functional groups and metabolic activities. Research can provide basal data on the distribution, diversity and functions of methane-oxidizing bacteria and ammonia-oxidizing bacteria from Mongolian plateau lakes and wetlands, and also provide the scientific basis to explore solutions to environmental issues by the eutrophication of lakes in Northern China and agricultural pollution. The Wuliangsuhai prairie lake is located in the west part of Inner Mongolia, China. It is spanning from 108°43E to 108°57′E, and from 40°47′N to 41°03′N, with an area of approximately 293 km2 . The water depth is approximately 1m, with a maximum water depth of 3 m. The region of the Wuliangsuhai Lake is characterized by a temperate, continental monsoon climate. The average annual temperature is 6.7ºC. The average annual precipitation is 224 mm, of which 60% is summer rainfall (July-August).
