Complete 16S rRNA gene (V1-V9) sequencing in vineyard topsoil and young leaf phyllosphere

As part of the PRJEB79310 project, which focuses on bacterial 16S and fungal ITS metabarcoding of topsoil, leaf phyllosphere, and endosphere in grapevine plots with high and low susceptibility to downy mildew, we also sequenced the entire 16S rRNA gene (V1-V9) using PacBio HiFi technology to improve the taxonomic characterization of bacterial communities. We created pooled DNA samples from different microbial habitats, specifically the soil and leaf phyllosphere, by mixing DNA from two individual samples collected from each of the 10 vineyard plots sampled in 2022. Each plot provided 2 µl of DNA per sample, resulting in a combined DNA pool for each habitat type sampled in 2022.
