Comparative analyze at the transcriptomic level 1) of Venturia pyrina pear host resistance via the major apple resistance gene Rvi6, in Rvi6 overexpressing transgenic pear versus ‘conference’ susceptible variety; 2) of Venturia inaequalis pear nonhost resistance, in ‘Conference’ variety, 24 and 72 hours post inoculation. For host resistance analyses, sampling concerned the youngest expanded leaf of each plant labeled the day of the inoculation with Venturia pyrina VP102 strain. Each sample is a pool of leaves from three different plants. Two biological repeats (genotype x treatment x time) were collected during two independent scab inoculation tests. Leaf samples taken just before inoculation (T0) and at 8, 24 and 72 hours post inoculation of these two biological repeats are used to perform transcriptomics analyses. At each time, the ‘Conference’ variety is used as a susceptible reference and compare to the resistant Rvi6 overexpressing transgenic line 60AU. For nonhost resistance analyses, sampling concerned the youngest expanded leaf of each ‘Conference’ plant labeled the day of the inoculation with Venturia inaequalis EUB04 strain. Each sample is a pool of leaves from three different plants and two biological repeats (n=2) have been made by condition (genotype x treatment x time). Leaf samples taken just before inoculation (T0) and at 24 and 72 hours post inoculation are used to perform transcriptomics analyses: T0 versus 24hpi and T0 versus 72 hpi.
