Long non-coding RNA (LncRNA) is a significant epigenetic regulatory factor, and its allele-specific expression (ASE) may play an important role in mental disorders. Therefore, clarifying the relationship between allele-specific expression (ASE) of lncRNA and mental disorders is of great significance for an in-depth understanding of the molecular mechanism of mental disorders. We here report that ASE-driven allelic shift in LINC02449 in Bipolar disorder (BD) and schizophrenia (SZ) contributes to behavioral and synaptic abnormalities in mice. We found that when the alternative allele of LINC02449 was overexpressed in cells and mouse brain regions, the expression of differentially expressed genes would be significantly enriched in mental disorders, mediating enhanced synaptic excitability transmission. Taken together, our data supported that LINC02449 overexpression, driven by the rs149707223 alternative allele, affects the disease susceptibility of BD/SZ. RNA-seq profiling of over-expression the rs149707223 refence and alternative allele of LINC02449, and orresponding empty vector in SK-N-SN cells.
