Blood immunomap for prediction of responses to aPD1 immunotherapy in metastatic non-small cell lung cancer

This repository contains mass cytometry (CyTOF) data generated as part of the study: "Immune profiling of non-small cell lung cancer (NSCLC) patients undergoing immune checkpoint inhibitor therapy reveals enrichment of CD8⁺PD-L1⁺ T cells in non-responders" ( https://www.sciencedirect.com/science/article/pii/S258900422501065X ). In this study, we performed high-dimensional mass cytometry to construct a peripheral blood immunomap in NSCLC patients with metastatic disease receiving anti-PD-1 immunotherapy. Analysis of responders versus non-responders revealed an expansion of CD8⁺PD-L1⁺ T cells in non-responding individuals. These findings suggest a potential role for this population as a non-invasive biomarker of resistance to immune checkpoint blockade. Contents 1. Mass Cytometry (CyTOF) data Pre-processed CyTOF data files (cleaned single cell events with gaussian parameters and live/dead exclusion) (FCS format). Sample metadata with clinical response annotations (md, excel file). Antibody panel used for the analysis (annotated markers/metal tags) (panel, excel file). 2. Imaging Mass Cytometry (IMC) data Raw IMC files (mcd): - 5269a.mcd (sample_1): includes ROIs 002 and 003. - 6362a.mcd (sample_2): includes ROIs 001, 003, 004 and 005. IMC antibody panel used for tissue staining listing metal tags and target markers (excel file). Image metadata detailing ROI selection and annotations (excel file). Usage These data can be used for immune profiling, clustering analyses, and identification of immune cell subsets associated with therapy response in NSCLC. This dataset supports a research article currently in press. The DOI of the associated publication will be added upon release.
