Eukaryotic diversity in biofilms from a drinking water distribution network

Eukaryotic diversity in biofilm samples from a drinking water distribution network in Riga, Latvia was surveyed by means of sequencing 18S metabarcodes. 42 libraries of 18S amplicons were prepared with 574*f and 1132r primers from Hugerth et al. (2014) and sequenced on Illumina MiSeq. The amplicons include V4 and V5 regions of eukaryotic 18S rRNA gene. One sample is from a pipe of cold water and the rest of samples are from hot water pipes. Four samples were added later where V4 region of the 18S library was amplified with TAReuk454FWD1 and TAReukREV3 primers from Stoeck et al. (2010). Some notes: The first 42 samples show a lot of unspecific amplification and generally low sequence quality. There are some mammalian 18S sequences in the data because of the BSA that was added to PCR in order to overcome inhibition.
