Single cell RNA sequencing of embryonal carcinoma cell lines 2102EP-lenti-MPHv2, GCT27-lentiMPHv2 and NCCIT-lenti-MPHv2 cells after yolk-sac tumor-like differentiation in vitro. Stable MPHv2+ embryonal carcinoma cells were genereted by infection with lentiviruses (Lenti-MPHv2 plasmind and a hygromycin resistance cassette). This hygromycin resistance was used to select the Lenti-MPHv2+ cells. All three embryonal carcinoma cell lines (2102EP-, GCT27-,NCCIT-lenti-MPHv2) were differentiated by inducing endogenous SOX17 expression (by SOX17-lenti-SAMv2-lentiviruses) and treatment with recombinant Activin-A (100 ng/ml), CHIRON (3 µM), recombinant FGF2 (25 ng/ml) and recombinant Heparin (1 µg/ml). The treatment with the recombinant protein mixture was repeated every two days for eight days in total. Then, cells were isolated and three replicates of each cell line were pooled and sequenced.
