H1299 and H1299 HMGA2 KO cells were treated with 2μM SN38 for 48h. Cells from both samples were processed for PFGE. The DNA fragment smear ranging from 30kb up to 150kb was excised after ethidium bromide staining. ZymoClean large fragment DNA recovery kit was used for DNA extraction. As controls, genomic DNA from H1299 and KO cells were extracted using DNeasy Blood & Tissue Kit (Qiagen), and submitted for sequencing.
