This study aims to elucidate the molecular mechanisms governing the genome-wide deposition of the repressive histone mark H3K9me3 in Caenorhabditis elegans embryos. By identifying and characterizing the cofactors SECP-1 and SECP-2, which form a complex with the methyltransferase SET-25, we reveal how this complex targets repetitive DNA sequences to establish heterochromatin domains. Understanding this process sheds light on the epigenetic regulation of gene silencing and heterochromatin organization, which are critical for development and heritable gene expression patterns.
