Human fetal lung tip cells were purifed from developing human lungs at pseudoglandular and canalicular stages and in vitro cultured; pseudoglandular organoids and LinPOS organoids, respectively. Alveolar organoids were differentiated from LinPOS organoids in the alveolar differentiation condition. Differential NKX2.1 binding patterns in each organoid were profiled by targeted DamID-seq. Overall design: 1) CD44+ tip population from pseudoglandular organoids, 2) CD36+CD44+ population from LinPOS organoids, 3) Alveolar organoids (three biological replicates for each cell type)
