Gene expression analysis of T-cell leukemia/lymphoma associated mutations of PLCG1 in Hut78 cells

The most frequently mutated gene PLCG1 was identified in Adlut T-cell leukemia/lymphoma patients. Here, we employed bulk RNA-seq analysis to clariy regulated genes and signaling pathways of PLCG1 mutations in Hut78 cells. We further defined unique genes and pathways for PLCG1 mutation by comparing to T cell receptor stimulation group. PLCG1-GFP wild type (WT), R48W, S345F, and D1165H mutations were respectally introduced into human T-cell lymphoma Hut78 cell line by lentivirus. Flow cytometry was used to sort satble expression cell lines using GFP channel. T cell receptor stimulation group: PLCG1 wild type Hut78 cells were activated by anti-CD3/CD28 antibodies for 3 days.
