In order to determine whether the Hopx protein plays a functional role in regulating fate determination of cortical NPs, we constructed a mutant mouse strain using CRISPR-Cas9 strategy. The mutated Hopx contained an extra nucleotide after the normally 18th base of the coding sequence and thus resulted in a frame shift. We then analyzed the differential expressed genes (DEGs) between wild-type and mutant mice at E16.5.
