Genotyping of cowpea accessions conserved at National Genebank, ICAR-National Bureau of Plant Genetic Resources, New Delhi, using the ddRAD seq approach

Cowpea accessions from the National Genebank, ICAR-NBPGR, were field-grown and morphologically characterised. Seeds from the single plant progeny were used for DNA isolation and genotyping. The ddRAD-seq approach was followed for genotyping. For library preparation, genomic DNA was subjected to double digestion using Sphl and MluCI restriction enzymes. High-throughput sequencing was carried out using Illumina TrueSeq chemistry on the Illumina HiSeq X platform.
