Circular extrachromosomal DNA promotes accessible chromatin and high oncogene expression

The brief ATAC-see method for metaphase chromosome spread is as follow: metaphase sample was prepared onto a 1-mm coverslip and incubated with 50 nM of ATTO-590 transposome under 37°C for 30 min in the dark. After washed twice by 2× SSC with 0.01% SDS for 15 min, and once by 2× SSC with 0.2% Tween-20 for 15 min, sample was subjected to FISH procedures, and finally stained by 1 µg/ml DAPI and mount with VECTASHIELD antifade mounting media (Vector Laboratories).<br>
