Transcriptome changes in 2,3-butanedione induced bronchiolitis obliterans in rats

Bronchiolitis obliterans (BO) is a destructive fibrotic lung disease, which can be partly induced by 2,3-butanedione (also known as diacetyl, DA). However, the mechanism underlying the effects of DA on BO is not clear. In the present study, a bioinformatics analysis was performed using DA-treated or untreated lung tissues of rats. Overall design: Male Sprague Dawley rats (age, 8 weeks; weight, 300 g) received 1 week of environmental adaptation, and had ad libitum access to water and food. The rats were randomly divided into the following two groups: i) Control group, intratracheal instillation of aseptic distilled water; ii) DA group, intratracheal instillation of 125 mg/kg DA (Macklin Inc., Shanghai, China). Rats in the DA group accepted DA intratracheal instillation once a day for 7 days. Twenty-four hours after the last intratracheal instillation, the rats were sacrificed and lung tissues with bronchi were collected. We then performed gene expression profiling analysis using data obtained from RNA-seq of six samples per group.
