N-acetyltransferase 10 (NAT10)-mediated N4-acetylcytidine (ac4C) modification is crucial for mRNA stability and translation efficiency, yet the underlying function in mammalian preimplantation embryos remains unclear. Overall design: RNA-sequencing of fifty dsGFP- or dsNat10-injected morulae were performed by Beijing Genomics Institute (Shenzhen, China). The library preparation was performed following the SMARTseq2 protocol. After library construction, all samples were sequenced on DNBSEQ platform with paired-end. The raw reads were trimed using trim_galore. The clean reads were mapped to mouse genome (GRCm38) using HISAT with default parameter. The reads of each gene were counted by Htseq-count.
