The expression of 88 target genes and three reference genes (Pgk1, Rpl13a and Sdha) was analyzed in singlets using a RealTime ready Custom Panel 384 – 96+ with Light Cycler 480 Probes Master on a LightCycler480 (Roche) using the following conditions: i) 95 °C for 10 min, ii) 45 cycles of 95 °C for 10 s, 60 °C for 30 s, 72 °C for 1 s co-terminated by a plate read.
AAV2 = cells treated with AAV-U6-Diras2_2-hSyn::mCherry.3xFLAG-WPRE
AAV4 = cells treated with AAV-U6-Diras2_1-hSyn::mCherry.3xFLAG-WPRE
Scr = cells treated with AAV-U6-Scrambled-hSyn::mCherry.3xFLAG-WPRE
Unb = untreated cells
本研究对88个目标基因及三个参考基因（Pgk1、Rpl13a和Sdha）的表达进行了分析，分析采用RealTime ready Custom Panel 384-96+，结合LightCycler 480 Probes Master试剂在LightCycler480（罗氏）仪器上进行，具体操作条件如下：i）95°C加热10分钟，ii）进行45个循环，每个循环包括95°C加热10秒、60°C加热30秒、72°C加热1秒，并辅以板式读数终止。AAV2组为经AAV-U6-Diras2_2-hSyn::mCherry.3xFLAG-WPRE处理的细胞，AAV4组为经AAV-U6-Diras2_1-hSyn::mCherry.3xFLAG-WPRE处理的细胞，Scr组为经AAV-U6-Scrambled-hSyn::mCherry.3xFLAG-WPRE处理的细胞，Unb组为未处理的细胞。
