This study identifies where RNA polymerase pauses on human DNA, and determines how RNA polymerase pausing is regulated. RNA polymerase pausing is increasingly recognized as a key regulatory step in transcription that affects gene expression. We mapped the locations of RNA polymerases in human cells by the precision nuclear run-on assay (PRO-seq) and located the NELF and DSIF complexes by ChIP-seq. The data set contains technical replicates of PRO-seq from foreskin fibroblasts, NELFA and SPT5 ChIP-seq from foreskin fibroblasts, and PRO-seq from a renal proximal tubule cell line (HK-2).
