This study aimed to evaluate MPS data that was generated with the ForenSeq™ DNA Signature Prep kit to characterise 24 Y-STRs and 94 iiSNVs in South Africans. Data from all participants (380 males, 240 females) were included in the iiSNVs analysis, while the Y-STR analysis focused on the biological male subset. Y-STR sequences were named using recommended nomenclature, and a shorthand naming convention was explored to name iiSNV sequences. Haplotype and allele frequency data were generated for the Y-STRs and iiSNVs, respectively, and population parameters were assessed. This dataset contains the naming sheets, as well as the iiSNV and Y-STR frequencies and parameters generated in this study, for the South African population.
