Despite its potential as a carbon source for low-emission industrial biotechnology, information on the physiology and energetics of ethanol-grown yeast cultures is limited when compared to the wealth of knowledge on sugar-based growth and productivity of yeast. It is currently unclear to what extent this sugar-based knowledge is transferable to a future where ethanol is a predominant substrate, rather than product, of yeast-based industrial biotechnology. In this thesis, the physiology and energetics of different yeast species, grown on ethanol as sole carbon and energy source, are explored to understand and optimize key performance parameters of yeast cell factories for ethanol-fueled industrial biotechnology.<br>In Chapter 1, the Introduction, ethanol metabolism in yeast has been investigated through thermodynamic and metabolic flux analysis, for which the data has been deposited here. In Chapter 6, the Outlook, preliminary data on a rational metabolic engineering approach regarding acetyl-CoA synthesis is discussed, for which the data has been deposited here.
